Screening of the attachment base of artificial breeding of Sargassum fusiformis

An important step in the cultivation of artificial seedlings of sargassum fusiforme.setch is to screen the attachment bases of the seedlings. Attachment bases are related to the success or failure of nursery seedlings. Attachment bases can allow seedlings to adhere and be reasonably distributed and grow well in space. Appropriate attachments should not be rotted, easy to handle, easy to obtain, non-toxic, low cost, and the seedlings should grow well on the attachment base.
l Test method
1.1 The cultivation, processing, and harvesting of a variety of Laiyang Laiqie were harvested from Dongtouhai District in early June. The planting trusses were divided into two sections. In 1999, 100 kilograms of fresh Levin were used, and the area of 130 m2 was selected; in 2000, fresh Ley 60 Kilograms, the area of 100 m2. Choose a good quality of mature Lai, male and female ratio of 2:1. About 3 to 8 hours of dried vegetables, the dehydration rate is 65% to 75%. 200 mesh sieve picking seedlings, a total of completed picking area
230 square meters.
1.2 Attachment base tests include five attachment base materials such as Dacron cloth, chemical fiber cloth, vinylon rope, bamboo curtain, and reef. Except reefs are collected from the sea area, the rest are provided by the Fenghua kelp seedling plant, and are soaked for 60 days. Change the water every 10 to 15 days 1 times, armpit hair treatment. The soaking treatment of bamboo curtains is more stringent.
1.3 Indoor culture and management of seedlings are divided into three periods. The natural light and temperature of the single-cell algae cultivation room are used at the initial stage. The seawater used for culture is treated with bleaching powder and 1/2 of MAV nutrient solution is added. In the mid-season cultivation in the kelp nursery room, dimming, natural water temperature, plus MAV nutrient solution. Later in the kelp nursery room culture, dimming, low temperature water with kelp breeding seedlings to reduce the temperature. The average temperature of seawater is 20°C, and the salinity is 29.8 ‰, pH 8.2.
1.4 Outdoor Cultivation and Management of Seedlings Before and after the mid-November, the sea was raised and the trusses in the wind gate sea area and Dongtou sea area of Xiangshan were stocked. During this period, the average temperature of the seawater is 12°C to 22°C, the salinity is about 30°F, and the pH is 8.0 to 8.3.
1.5 Indicators a. Length: Measure the average length (micrometers) of the length of 20 seedlings from the base of the rhizoid to the top of the stem; b. Growth rate: The growth rate is measured with the length as an indicator; c. Adhesion rate: Adhesion rate = the number of young spores attached / the total number of young spores x l00%; d. Seedling-attached rate: Seedling-attached rate=number of seedlings attached to the sea/total number of seedlings×100%.

2.1 Differences in the effect of different attachment groups on growth of seedlings The attachment of five attachment groups, ie, good fabric, chemical fiber cloth, vinylon rope, bamboo curtain, and reef, showed that the young sporophyte had different attachments to the attachment base in the growth of hijiki seedlings. Growth rate, see Table 1. For the first 20 days, the growth rate of Dacron cloth, chemical fiber cloth, and Vinylon ropes was relatively large, followed by bamboo curtains and the reefs. The 60-day growth rate is basically the same, and the length of the chemical fiber fabrics, Vinylon ropes, and bamboo curtains are almost the same. Reefs are relatively small. The growth rate at 120 days is even smaller for different attachment groups. The difference in the length of growth of seedlings between different attachments may be related to the retroreflectivity of the attachment. It can be considered that there are no obvious differences in the growth of the seedlings of the Yangmule seedlings, such as Dacron cloth, chemical fiber cloth, Vinylon rope, bamboo curtain, and reefs. From the standpoint of growth, it can be used as the attachment base for nursery.

2.2 Differences in the effects of different attachment groups on the attachment rate of young sporophytes The five attachment groups, ie, good fabric, chemical fiber cloth, vinylon rope, bamboo curtain, and reef, were attached to the young larval sporozoites of the ovine larvae, showing the presence of young sporophytes on the attachment. Base has different adhesion rates, see Table 2. In the first 20 days, the adhesion rate was relatively high. Indeed, the growth rate of good fabrics, chemical fiber cloths, and vinylon ropes was relatively large. The bamboo curtain was the second and the reefs were the smallest. The 60-day adhesion rate was indeed good fabric, chemical fiber cloth, vinylon rope, and bamboo blinds. Basically the same, the length is also close. Reefs are relatively small. 120 days adhesion rate for different attachments. The difference between the bases is even smaller. During the incubation process, the relationship between the formation and growth of attachment and pseudo-roots was observed. The number of pseudo-roots of larvae is basically six or more, with an average of about 11 and a maximum of 25. The length of the fake root is basically close. There is little difference between the different attachment groups. The initial attachment of young sporophytes was not different. In the process of seedling cultivation, spraying with 0.6 kg/cm2 of water pressure had certain differences in the formation, growth and attachment of young roots. Bad cloth is indeed good, followed by chemical fiber cloth, and reef is the worst. It can be considered that there are certain differences in the attachment of five attachment bases such as Dacron cloth, chemical fiber cloth, Vinylon rope, bamboo curtain, and reef to the young seedlings. In consideration of the adhesion of young sporophytes, bamboo and curtains can be used as the attachment base for seedlings. Vinylon rope can be used as another choice for nursery attachment. In addition, bamboo curtains are dense and hard, or because of other reasons, false roots are difficult to attach firmly.

Reef 65.5 52.1 25.9

2.3 Effect of different attachments on seedling emergence rate in the sea under seedlings After the attachment base has a certain number of seedlings attached, when the temperature steadily drops below 22°C (from late October to mid-November), certain sea areas are selected for cultivation. The effects of attachment bases on the seedling-bearing rate in the sea under seedlings are shown in Table 3. There are no obvious differences in the growth of seedlings for different attachment groups. For the seedling-attached rate, bamboo curtains are the best, Venetian ropes are next, and indeed cloths, chemical fiber cloths, The seedling rate of the reef is the worst. At the same time, the experiment also found that when the water layer below the transparency was cultivated, it grew slowly and did not even grow. However, diatoms, green algae, and red algae are more likely to adhere when the surface layer is raised, which affects the growth of seedlings. Considering the effect of the attachment base on the seedling emergence rate under the seedlings, the attachment base is the best bamboo screen, followed by the vinylon rope.

Table 3, Effect of Attachment on Seedlings and Growth of Seedlings in the Sea Under the Seedlings

120 days before the sea
Number of seedlings (tree/cm2) Length (mm) Number of seedlings (tree/cm2) Percentage of seedlings attached (%) Length (mm)
Bamboo blind 5.83 8.7 4.36 74.8 12.9
Venetian Rope 4.34 8.5 2.19 50.5 12.6
Indeed, 2.95 8.5 1.03 34.9 12.2
Chemical fiber cloth 3.09 8.6 1.21 39.1 12.5
Reef 2.24 8.5 0.83 37.1 12.4

3 Conclusion The indoor cultivation of composite attachments and the cultivation of the limulus, bamboo curtains can be used as the attachment base for the artificial breeding of Sargassum. In addition, Vinylon rope is also an option. Indeed, good cloth, chemical fiber cloth and reefs have some deficiencies as attachment bases for productive seedlings.
