Introduction to fully automated DNA library construction technology

In the second-generation sequencing, the construction of the gene library is a very time-consuming and labor-intensive task, requiring skilled scientific and technical personnel to maintain high attention throughout the operation process, and spend a lot of time and effort to complete the database construction work. Therefore, most laboratories have the need to automate library construction efforts to save manpower.

Should be the need to build library automation, Pema. PrimaDiag has developed the ACSIA NGSLibPrep automatic database platform, which is highly accurate and reproducible. It uses the innovative patented APIM (Inductive Magnetization Attraction Technology) to process the magnetic beads and generate magnetic force when power is turned on or off according to demand. Demagnetization, temperature control is accurate to 0.1 °C, and the oscillation speed is adjustable from 200 to 3000 rpm.

The optimized operating procedure allows the entire library to be completed with a minimum number of tips, saving significant reagent and consumable costs. For example, the construction of 48 DNA libraries requires only 3 cartridges, while the number of guns used in manual operations can reach as many as 14 cartridges.

Comparing the fully automated and artificially established libraries of ACSIA, the DNA fragments were 338 bp and 340 bp, respectively, and the results were very similar. The library quality obtained by using ACSIA was nearly 30% higher than that of artificial libraries.
