Color calla lily tissue culture rapid propagation production technology

Colored Calla Liriodendron calla, perennial herb bulb flower. It is a flower that has a large variety of colors, and is a variety of flowers that are popular in the flower market. The large flames of red, yellow, pink, and orange are bright and colorful. Most of the green leaves of calla lily with white spots or stripes on the moon can be used as a leaf material with good ornamental value. Traditionally, color calla lily is propagated using the method of dividing tubers. Its propagation coefficient is low and multiple generations reproduce asexually, often causing degradation of varieties. In 1999, we introduced four different colors of calla lily from China Academy of Agricultural Sciences for the research of tissue culture and rapid propagation technology. After two years of experiments, we have produced a large number of tissue culture seedlings in the greenhouse in the short term. Transplanting survived and the plants grew strong. The specific production process of tissue culture seedlings is as follows:
First, aseptic seedlings cut test species: colored calla lily sterile test tube plant from the Chinese Academy of Agricultural Sciences, Institute of Vegetables and Flowers. A total of 8 varieties (colors), 50 bottles, 4 to 6 buds per bottle, shoots 3 to 4 cm high, light green color, seedlings growth potential in general. Place the seedling bottle 2m away from the UV source, turn on the UV lamp for 60 minutes, and then place it on a culture shelf in the culture chamber for 5 days. Prepare for cutting. The bottle seedlings were placed under aseptic conditions and carefully removed from the bottles. According to the growth status of the seedlings, long leaves (up to 2 cm above the leaves) were cut off, and the base leaflets of the tubers were preserved, and the colored calla tubers were cut longitudinally, each small piece. Two to three leaflets were retained and inoculated onto MS medium supplemented with different hormones. Each flask received 3 to 4 small pieces. The composition of the culture medium was:
Second, proliferation of sterile seedlings The cutting seedlings were placed on a culture rack in a culture room. The temperature of the culture room was controlled at 25° C. and 2° C., the light intensity was 2000 lux, and the light irradiation time was 12 hours/day. After 1 week of culture, the buds began to grow from 1:10 to 1:20, and small buds began to sprout. After 2 to 3 weeks of culture, clusters of small buds begin to form around the buds. After 4 to 5 weeks of culture, the leaves are elongated. The height of the leaves is as high as 3 to 5 cm and the minimum is less than 1 cm. Dense and germinating small buds are still visible on the tubers of dense leaflets. Tests have shown that the colored calla lily seedlings, in addition to different concentrations of 6BA and NAA medium, can be a large number of proliferation, proliferation rate in the 25-40 days up to 1:10 to 1:20. All kinds of calla were able to induce sprouts in the appropriate medium, but there were certain differences in the number of buds formed between the varieties and the growth rate of buds. The most varieties can be cut more than 20 bottles per bottle, the least can also cut more than 10 bottles.
Third, rooted seedlings cultured sterile seedlings in the culture room after 30 days of culture, the height of 2-3cm above the sterile seedlings, transferred to the rooting medium for rooting strong seedlings culture, 7 to 15 days later, there are more The root system is generated. Tests showed that in the MS medium plus NAA 0.3mg/L; IAA 0.2mg/L; NAA0.3+IAA 0.2mg/L of different hormone components, MS plus IAA 0.2mg/L in the rooting medium The highest seedling rooting rate was 96%, and the root system was short and thick, and the roots were evenly distributed. The roots were white, and each plant had 4 to 6 roots. The leaves were dark green with strong growth. In the rooting medium supplemented with NAA 0.2 mg/L, the leaves were dark green and the roots grew slowly. After two months of culture in the bottle, the rooted shoots still had no obvious tubers, but the roots grew rapidly and spread the bottom of the bottle. The leaves of the young seedlings were yellow, and with the prolonged cultivation time, they gradually died of dryness. Fourth, tissue culture seedling transplanting technology rooted seedlings carefully removed from the bottle, wash away the media, according to the spacing of 5cm6cm spacing transplanted in the peat soil plus half perlite bed. The seedbed is 1.2m wide and the length depends on the amount of transplanted seedlings. Transplanting seedlings take a small shed to moisturize, at noon every day, pay attention to open the ventilation at both ends of the tent, pay attention to moisturizing within one month of transplanting seedlings are not regularly sprayed one-thousands of elemental diluent to supplement nutrition. The transplanted seedlings began to grow rapidly after 1 month, at which time the light was increased and the leaves were fertilized. The seedlings grew vigorously.
