Shanghai experts confirmed - streptococcal DNA can cause psoriasis

Shanghai Jiejin Hospital Zheng Jie and other studies have confirmed that streptococcal DNA components may lead to the onset of psoriasis, the research paper "bacterial DNA components in streptococcal antigens enhance the proliferation of peripheral blood mononuclear cells (PBMC) in patients with psoriasis vulgaris and Activation was published online July 16, 2009 in J Investigat Dermatol. By studying the effects of streptococcal DNA on immune cell proliferation, activation, and changes in various cytokine secretion levels in patients with psoriasis, Zheng Jie et al found that PBMC in patients with psoriasis were stimulated by streptococcal antigen (SA) compared with healthy people. After that, it has a stronger proliferative response ability. The researchers used DNase-I to remove the nucleic acid component of the streptococcal antigen to obtain a nucleic acid component-removing streptococcal antigen (non-NASA). The proliferative effect after stimulation of PBMC with non-NASA was significantly lower than that of the SA-stimulated group. Furthermore, compared to SA stimulation, non-NASA significantly reduced CD69 levels (including T cells expressing skin homing lymphocyte-associated antigen) and PBMC-secreting alpha interferon (IFN-α) levels. However, for patients with non-NASA stimulation, the level of T cell activation and IFN-α secretion decreased. If synthetic CpG-A was added to the stimulus, it can be restored to the level of SA stimulation alone. The effect of CpG-A is positive. It is a DNA component that mimics streptococcus. Compared with the two groups of stimulation, the B cell surface activation marker CD86 expression level and γ-interferon (IFN-γ) and tumor necrosis factor-α (TNF-α) secretion did not change significantly; but the artificial CpG was added at the same time of stimulation. The expression level of CD86 was significantly increased after -B. By revealing the biological functions of streptococcal antigens (especially streptococcal DNA), this study found that non-NASA can significantly reduce PBMC proliferation and T cell activation in patients with psoriasis, suggesting that streptococcal DNA can assist in the activation of pathogenicity.
