Please carefully check the chiller, air compressor, reactor, inlet and outlet pipes, and air pipe before operation. Fill in the instruction manual.
Power on: Turn on the switch on other units first, then turn on the switch on the back of the unit.
Shutdown: first adjust the parameter status of all units on the controller to off, turn off the air intake switch and turn it off;
Turn off the switch on the back of the controller and turn off the switch on the other unit.

Sterilization operation
1. Remove the motor and lay it flat on the table. A black protective cap is placed over the top of the can.
2. Remove the temperature sensor from the tank. The sensor does not need to be sterilized.
3. Remove the PH cable and cover with a red protective cap; insert the PH electrode to the end. Be sure to tighten the upper and lower fixing nuts of the electrode. The PH electrode should be calibrated before sterilization. PH electrode calibration: After connecting the cable, unscrew the fixing nut (*) of the electrode, remove the PH electrode and rinse it with deionized water. Gently use the tissue to absorb the water and then put the standard solution 4.01. In the middle calibration, wait for the data on the controller to stabilize and input 4.01 at set zero (no matter the weak difference of the actual value); after the input is closed, take out the electrode from the standard solution, rinse it, blot it, put it into the standard solution of 7.00, etc. After the data on the controller is stable, enter 7.00 at the set span (regardless of the slight difference in actual values). Do not turn off the controller after the PH electrode is calibrated.
4. Remove the DO cable and place the black cable cap on the DO cable interface with a black cap. The DO electrode is calibrated after sterilization. Insert the DO electrode to the end and tighten the upper and lower fixing nuts of the electrode. Before sterilizing, pay attention to whether the protective liquid inside the dissolved oxygen electrode is still there. If it is not, add it.
5. Remove the defoaming or liquid level cable, insert the defoaming or liquid level cable into the end, and after positioning to the required height after sterilization, do not press it down. The height of the defoaming electrode is about 1.5 cm from the liquid surface.
6. Remove the inlet and outlet pipes on the fermenter and the condenser. At the same time, remove the joints of the removed water pipes with clips to prevent water from flowing out.
7. The ends of the filter on the fermenter should be clipped with clips. The outlet of the filter should be wrapped with tin foil. The ends of all filters should be clamped with clips. After the fermentation is finished, check whether the filter is wetted, and if so, use the suction ball to blow out the liquid.
8. Clamp the silicone tube on the harvest tube with an iron clip to prevent running in the sterilizer. When the harvest tube is not in use, it must be stuck and not removed.
9. Before sterilizing, loosen the six fixing nuts of the fermenter and the other nuts must be tightened! Immediately after opening the pressure cooker after sterilization, tighten it, and wear gloves to prevent burns.
10. Do not sterilize the acid in the pressure cooker. Wrap the filter on the empty acid bottle with tin foil and loosen the cap. After sterilization, the foil paper on the foil filter is removed as an outlet. (Note that the silicone tube connected to the acid bottle should not be stuck); the lye bottle and the silicone tube can be sterilized together (the silicone tube connected to the lye bottle is stuck).
11. Remove the electric heating jacket and lay it on the table top.
12. Adjust the position of the stirring blade before sterilization. The bottom stirring blade should be as close as possible to the air distributor, and the upper position should be adjusted to the middle position of the added fermentation liquid.
13. The position of the sampler should not be close to the position of the baffle, as close as possible to the middle position.
14. Remove the suction ball from the sampler before sterilization. Do not rely on the inner wall of the pressure cooker for all silicone tubing.

After sterilization
1. Tighten the six loose fixing nuts and the acid-base cap.
2. Remove the clips from the foil and the silicone tube wrapped in the filter.
3. Install the motor. If the lubricant on the inside of the motor is dry, apply a proper amount of Vaseline.
4. Connect the DO electrode after connecting the dissolved oxygen cable: after the sterilization, connect the cable of the DO electrode and input 0 at the set zero; control the air flow of the reactor to 1L/min, and Agitation to 100r/ Min overnight, open in the morning at the set span, enter 100 and press the enter key.
5. When the temperature drops to the temperature required for fermentation, connect each inlet and outlet pipe, PH cable, and defoaming cable.
6. Before placing the temperature electrode, add glycerin to the inlet of the temperature electrode, insert the electrode and continue to drop to the end. Note that the temperature electrode is inserted all the way.
7. Check the output pressure of the air compressor, which should be in the range of 0.7~1.0kg/cm3.
8. The silicone tube of the defoamer should be installed on the pump in order from bottom to top.

Cold water machine working temperature setting
Start up: connect the inlet and outlet pipes → turn on the main power supply → perform temperature design → turn on the circulation pump → start cooling
Shutdown: first turn off the cooling switch → turn off the circulating pump switch → close the inlet and outlet valves simultaneously → turn off the power
Temperature setting: Power on → Panel displays “PP0.0” → Press “Shift Key” and “Adjustment Key” to display “PP0.1”
→ Press the SET button to confirm → the panel displays the upper limit value, press the “Shift Key” and “Adjustment Key” to set the upper limit value → press the SET button to confirm → the panel displays the lower limit value, press the “Shift Key” And “Adjustment Key” to set the lower limit value → press the SET key to confirm → press the SET key again → press the SET key → the panel displays the end mark “---”, press the SET key to return to the measurement display, and the setting ends.

Prepare for vaccination after the above operation
Preparation for inoculation: Prepare iron baffle, lighter, alcohol spray can, inoculation ring, enamel cylinder container with 95% ethanol, pointed pliers, adjustable wrench, inoculating bottle.
Vaccination operation
1. Remove the hose near the inoculation port, enclose the iron baffle around the inoculation port, and then spray it with alcohol sprayer toward the inoculation port. (Note: If there is a flame that overflows the iron plate with alcohol, you need to blow it out.) The flame that overflows on both sides of the iron plate.)
2. Place the inoculation ring with alcohol and cover the inoculation port. Dip the wrench or pliers into the nut and unscrew the nut. Then clamp it with pliers, blow out the flame on the nut and pliers, and then put it in quickly. In a 95% ethanol enamel tank container.
3. Pour the inoculum bottle into the fermenter after it is pulled out in the flame. When finished, pliers are pulled out with pliers. The entire process ensures aseptic operation!

Dissolved oxygen setting: There are four cases of control of dissolved oxygen in fermentation mode: press the off key → press the control key → press the Auto key → setup → cascade → select the following four modes.
1. None state: The control of dissolved oxygen is mainly controlled by increasing the air flow rate and setting a fixed speed.
2. Agit state: The control of dissolved oxygen is mainly controlled by increasing the rotational speed. In this state, when the air flow rate remains unchanged, the dissolved oxygen increases the dissolved oxygen level by increasing the rotational speed as the cell consumption decreases. Until the speed reaches the set speed upper limit. This process is automatically controlled.
3, 02 Status: This state is mainly controlled by controlling the mixing percentage of oxygen and air. Dissolved oxygen increases as the percentage of oxygen increases. For automatic control of the process.
4, Agit / 02 state: the control of dissolved oxygen in this state is first increased by increasing the speed, when the speed reaches the set upper limit, if the actual dissolved oxygen value has not reached the required set value, then the speed Controlling at the upper limit, the amount of oxygen entering is automatically increased from small to large until it reaches the set dissolved oxygen value. This process is also an automatic control process.
Note: When using the Agit state and Agit/02 state, if the actual dissolved oxygen value is too different from the set value, make sure that the set stirring speed upper limit is not too large, otherwise the speed will increase in an instant. Too much shear and a lot of foam. In actual operation, the manual control of the air flow meter can be used to assist the operation.

Other control operations:
Select U1:1, after booting → press the use button → press the Fermentation or cell culture button of the U1~U4 unit to select the fermentation mode or the cell culture mode; press the scroll key continuously → control the single unit of U1~U4 separately.

Pump A: 1. Off→ press control → press manual → press setpoint to set the percentage (in 10 seconds, if you type 20%, turn 2 seconds, stop for 8 seconds: 100% means continuous operation) → Disp Hi and DispLo Set the same as above. The settings for PumpB and PumpC are the same as for PumpA.
2, off → press the control button → press the Antiforma button → wet on and wet off to adjust the level of the liquid level.
3. PumpC is not connected to the peristaltic pump for the time being; the peristaltic pump 3 is connected to the alkali, and the peristaltic pump 4 is connected to the acid.
PH value setting: 1.off→press the control key→press the auto key→press the setpoint key→press the set value and press the OK key→Disp Hi and Disp Lo values ​​as above.
dO2 value setting: Press the off button → press the control button → press the Auto button → press the setpoint button → enter the set value and press the OK button.
O2 flow setting: Press the control key → press the Auto key → press the setpoint key to enter the set percentage.
The operations of Unit2, Unit3, and Unit4 are the same as above.
